GT HSTaq (Hot Start Taq DNA Polymerase)

Hot Start Taq DNA Polymerase is a mixture of Taq DNA polymerase, either with antibody, chemically modified or an aptamer-based inhibitor. All these reversibly binds to the enzyme and/or inhibits polymerase activity below 45°C and chemically modified becomes more active as PCR progresses. 

Genetek Biopharma's HSTaq DNA Polymerase with 96 kDa molecular weight is a, non-antibody bound, heat stable Taq DNA polymerase in a proprietary formulation. It is a highly processive 5’ to 3' DNA polymerase, lacking 3'-5' exonuclease or proofreading activity. This recombinant enzyme reaches its peak enzymatic activity at 72 °C.

Each GT HSTaq DNA Pol package contains 10X Buffer and MgCl2. Mg+2 is required to increase the total enzymatic activity of the Taq polymerase. Therefore, MgCl2 is provided as a separate solution to be add to the 10X buffer. The optimal concentration of MgCl2 is important as it can increase the rate of non-specific binding during the amplification process. A concentration of 1.5 mM is usually sufficient for most PCR applications.

The GT HSTaq  is heat stable therefore it can withstand high temperature for much longer time than GT Taq DNA pol. Indeed its activation requires preheating for 5-20 min depending on the type of PCR. If you use it in kits or PCR with kits other than Genetek kits, please try to optimize preheating time required for enzyme activation. Preheating is usually carried out at 95 °C for about 15-20 min.
