The GT AZFScreen Plus v2 kit is an updated AZF kit to comply with the EAA/EMQA 2023 Best Practice Guideline. It is intended for the detection of Y chromosome microdeletions and Klinefelter Syndrome, the two most frequent genetic causes of infertility in men. This kit includes 36 markers (26 STS markers, 3 SD [Segmental Duplication] and 7 STR [Short Tandem Repeat]). Primer designing followed European Molecular Genetics Quality Network guidelines.
Infertility is regarded as a critical problem for most couples. It affects 15-20 % of the reproductive-aged couples worldwide. Male infertility contributes to about 20-30 % of these cases, out of which Y chromosome microdeletions cause 5-10 %. About 3 % of infertile men and 14 % of non-obstructive azoospermia men are affected by Klinefelter Syndrome.
The GT AZFScreen plus v2 kit having 7 X chromosome- specific STR markers as well as 3 segmental duplications can detect Klinefelter Syndrome (47, XXY) and other X or Y chromosome aneuploidies (e.g., 47, XXX, 48, XXXY, 48, XXYY, 48, XXXX, etc., though only those related to infertility is of our concern). The GT AZFScreen Plus v2 kit performance has been validated with extensive testing using Applied Biosystems™ 3500/xL and 3130/xl platforms for detection and analysis on more than 200 samples, either as normal, with Klinefelter Syndrome, deletions of AZFa, AZFb, and AZFc or any combination of these deletions on Y chromosome as well as other X chromosome aneuploidies. Compatibility
This kit is compatible with 5-dyes capillary electrophoresis system such as with Compact Spectrum CE System from Promega or ABI 3130/xl, 3500/xL , and SeqStudio Genetic Analyzers with either 30, 50, or 80 capillaries.
This kit can be used on various sources of DNA including DNA extracted from various sources or DNA obtained using any of extraction free (direct) materials from Genetek such as GT BLB (Blood Lysis Buffer), and filter papers (GT DBC or GT DBC Blue).
Kit Content
The kit contains all the necessary reagents and buffers for multiplex PCR (Multiplex Primer mix, PCR buffer, GT HSTaq DNA Pol in Box A). Also, the kit comes with the GT500 internal size standard as well as GTM5 v2 Matrix Standard for Spectral Calibration of CE systems.
